In the feed additive industry, finding safe and effective antibiotic alternatives remains a key focus. α-Glycerol monolaurate (α-GML), also known as α-monolaurin, is a natural medium-chain fatty acid monoglyceride. Recent research in swine, poultry, and aquatic animals has shown promising results in improving gut health, immune response, and antioxidant status. Swine: α-GML Reshapes Gut Microbiota and Promotes Colonization of Beneficial Bacteria

Sows received α-GML from late gestation through lactation. The trial included four groups: CON = 0, GML1 = 500 mg/kg, GML2 = 1,000 mg/kg, and GML3 = 2,000 mg/kg, with n = 6 per group.

Key findings in suckling piglets:

  • ✔ The abundance of Firmicutes and Lactobacillaceae increased significantly.
  • ✔ Potentially harmful bacterial families such as Enterobacteriaceae and Enterococcaceae decreased significantly (P < 0.05).
  • ✔ Proteobacteria showed a downward trend (P = 0.098).

These changes help maintain intestinal microbial balance and support immunity and healthy growth in piglets. The results suggest that maternal α-GML supplementation can support the transfer of beneficial microbiota from sow to piglet, helping piglets build a healthier gut environment early in life.

Poultry: α-GML Helps Enhance Immune Response and Defensive Capacity

A total of 480 one-day-old Ross 308 broilers were randomly assigned to four groups and fed diets containing 0, 0.25, 0.5, or 1.0 g/kg α-monolaurin for 33 days.

Key findings:

  • The 0.25 and 1.0 g/kg groups had significantly lower hepatic MDA (malondialdehyde) levels than the control group. This indicates reduced lipid peroxidation and improved antioxidant status.
  • After Newcastle disease virus (NDV) challenge, all supplemented groups had significantly higher antibody titers than the control in Week 2.
  • The 0.25 and 1.0 g/kg groups remained significantly higher in Week 3.

These results indicate that α-GML helps enhance immune response and defensive capacity in broilers, supporting poultry health and reducing reliance on antibiotics.

Aquatic Animals: α-GML Improves Intestinal Morphology in Nile Tilapia

α-GML also shows benefits in aquaculture. A study on Nile tilapia fed different levels of alpha-monolaurin reported significant improvements in intestinal morphology.

The trial used 240 juvenile Nile tilapia (initial weight 3.19 g), divided into four groups with three replicates per group and 20 fish per tank. Diets contained 0, 2, 4 or 6 g/kg α-GML for 70 days. α-GML significantly improved intestinal villus development and growth performance. Compared with the control, the 6 g/kg group showed an increase in villus length from 237 µm to 389 µm (+64%) and an 80% increase in villus width; final mean body weight increased by 23.6%, FCR decreased from 1.65 to 1.25, and survival increased from 97.5% to 100%. This was a high-dose research trial (villus length increased significantly at ≥4 g/kg, while villus width and goblet-cell number increased significantly from 2 g/kg; the original paper recommended a dietary inclusion level of 4 g/kg, and broken-line regression indicated an optimal FCR point of 3.2 g/kg) .
